The present invention provides a single-use culture apparatus and a culture method, in both of which bubbles formed on the liquid surface of a liquid culture medium rarely spill over an exhaust port of a culture vessel. The single-use culture apparatus (100) is provided with: a culture vessel (1) in which a liquid culture medium (7) can be enclosed; a submerged aeration means (13) which is arranged at a bottom part of the culture vessel (1) and enables the submerged aeration of the liquid culture medium (7) enclosed in culture vessel (1) with oxygen; an exhaust port (110) which is arranged at an upper part of the culture vessel (1) and through which a gas in the culture vessel (1) can be discharged; and a blocking sheet (4) which is arranged at a bottom part of the exhaust port (110) and can partition between the exhaust port (110) and the liquid surface of the liquid culture medium (7). The culture method is carried out in the single-use culture apparatus (100) and comprises enclosing a liquid culture medium (7) and cells in the culture vessel (1) and culturing the cells while carrying out submerged aeration of the liquid culture medium (7), wherein a fresh culture medium or an antifoaming agent is supplied onto the blocking sheet (4) during the culturing of the cells.
