The present invention relates to a peptide for inhibiting the phosphorylation of a 120^th threonine (T120) residue of testis-specific Y-like protein 5 (TSPYL5). More specifically, T120D and T120A-TSPYL5 genes, which are mutants of the T120 residue of TSPYL5, are prepared and transformed into cells to confirm influence by phosphorylation of the T120 residue. As a result, it has been confirmed that normal-type T120YL5 and T120D migrate into the nucleus, but TSPYL5 cannot migrate into the nucleus but is expressed only in the cytoplasm in T120A, and cannot bind with AKT; ubiquitination of the TSPYL5 is increased; and sumoylation is inhibited. Also, it has been confirmed that the expression of ALDH1-A1, -A3, CD44 genes and protein is reduced; the growth and metastasis of lung cancer cells are reduced; and sphere formation is reduced. On the basis thereof, a peptide consisting of the amino acid sequence of SEQ ID NO: 43 or SEQ ID NO: 44 of the present invention, which inhibits the phosphorylation of the T120 residue of TSPYL5, is designed, thereby being effectively used as a cancer cell growth or metastasis or cancer stem cell growth inhibitor composition.
