The present invention is a technology for increasing the content of lentinan by activating mitochondria of fragrant mushroom lentinus edodes using a special mixed composition for increasing a content of lentinan in fragrant mushroom lentinus edodes. A cultivation method of fragrant mushroom lentinus edodes for increasing the content of lentinan according to the present invention comprises the steps of: (A) preparing an oak log with a bark; (B) forming a seed hole in the log, and injecting seeds of fragrant mushroom lentinus edodes into the formed seed hole to inoculate the seeds of fragrant mushroom lentinus edodes; (C) performing an inoculated log management process by spraying water onto the log once every week in 15 days after laying down the log (bed log) having fragrant mushroom lentinus edodes injected thereinto under conditions of shade of 60 to 70% and light of 40 to 30%; (D) performing a process of setting up a seed culture-rooted log in which mycelia have been grown as much as 80% or more during a period of November to December, or a spring period that is the following February; (E) dissolving the mixed composition according to the present invention into water to produce a mixed composition solution; and (F) repeatedly spraying the produced mixed composition solution onto the bed log at intervals of 15 days until the harvest of fragrant mushroom lentinus edodes.
