The present invention is directed to Fc region-containing polypeptides exhibiting improved effector function due to altered degree of fucosylation, and methods of using such polypeptides to treat or prevent cancer and other diseases. The Fc region-containing polypeptide of the present invention is preferably an immunoglobulin (e.g., an antibody), wherein the Fc region comprises at least one amino acid substitution relative to the corresponding amino acid sequence of the wild-type Fc region, Is sufficient to attenuate and modulate the reduced binding to the enhanced binding and inhibitory Fc receptors for activated Fc receptors. The methods of the present invention are particularly well suited for use in the treatment of diseases where an enhanced efficacy of effector cell function mediated by Fc [gamma] R is required (e.g. cancer, infectious disease) or where inhibited effector cell responses mediated by Fc [gamma] Or ameliorate one or more symptoms associated with a disease, disorder or infection.
