In order to devise a method for searching a Chinese hamster ovary (CHO) cell hot spot region for recombinant gene high expression, the present invention establishes a CHO cell line expressing green fluorescent protein (GFP) by processing lentivirus including a CMV promoter and a GFP sequence, separates the CHO cell line which highly expresses the GFP among the CHO cell line into four sections through a flow cytometer having an automatic cell deposition device, selectively amplifies an insertion position of the lentivirus through a target PCR, and finds the hot spot region through sequence analysis of the corresponding gene insertion position through a next generation sequencing.
