The present invention provides methods and compositions for enhancing the specific enrichment of target sequences in nucleic acid libraries. Targeted hybridization probes can be used to reduce binding and / or capture of the target outside regions of the nucleic acid library in a targeted sequencing workflow. The off-target hybridization probe may be specific for a position known to generate an off-target sequence analysis reading for a particular set of hybridization probes.
