Embodiments of the present disclosure relate to the use of expression constructs that are embedded upstream of a transcriptional entity capable of expressing at least one polyA signal, e.g., within a 5'UTR for a transcript. In certain embodiments, the polyA signal is contained within a ligand binding capacity tamer and the binding of the ligand to the plaster, or lack thereof, describes the result for an expressible transcript. In a specific embodiment, the absence of a ligand produces an expressed transcript having a poly A in its 5 'UTR that is expressed and then degraded, while the presence of the ligand causes inhibition of degradation upon expression of an expressible transcript. More than one ligand binding aptamer can be present in the same expression construct.
