The present invention relates to a synergistic composition for anticancer activity of gemcitabine against pancreatic cancer containing 6-thioguanine as an active ingredient. According to the present invention, when 6-thioguanine and gemcitabine are administered in combination, pancreatic cancer cells are effectively inhibited, and since an excellent pancreatic cancer therapeutic effect is exhibited in an animal model of pancreatic cancer, the composition can be usefully used for prevention or treatment of the pancreatic cancer.
