The present invention relates to a culture medium containing high-purity and high-concentration exosome derived from mesenchymal stem cells, and a production method thereof. Specifically, according to the present invention, when exosomes are extracted from mesenchymal stem cells cultured in three dimensions, exosomes with uniform size can be obtained at a higher yield. Thus, the method can be useful as a high-purity and high-concentration exosome extraction method.
