The present invention relates to a nerve gas simulation agent analyzing method using an electrowetting digital fluid chip and a diol-functional group coated magnetic bead. The nerve gas simulation agent analyzing method using a digital fluid chip comprises: a step of loading a sample solution, in which a nerve gas simulation agent is dissolved, a magnetic bead solution which includes a magnetic bead, a solvent which is used for the sample solution, and an elution solvent in different storage electrode surfaces included in the digital fluid chip; a step of moving a droplet of the magnetic bead solution to a specific channel electrode of a plurality of channel electrodes included in the digital fluid chip, and changing a solvent included in a droplet positioned in the specific channel electrode into the solvent used for the sample solution; a step of mixing the droplet of the sample solution loaded in the digital fluid chip and the droplet positioned in the specific channel electrode to allow the nerve gas simulation agent to adhere to the magnetic bead surface; and a step of dissolving the nerve gas simulation agent adhered to the magnetic bead surface in the elution solution, and moving the elution solution, in which the nerve gas simulation agent is dissolved, to a concentration electrode included in the digital fluid chip. The nerve gas simulation analyzing method using a digital fluid chip can analyze a minute amount of the nerve gas simulation agent by using a magnet on the digital fluid chip through solid-phase extraction (SPE) using the magnetic bead.
