Disclosed is an optical gene biosensor. The optical gene biosensor includes: a substrate; a molecular beacon unit having oligonucleotide selectively binding to a target nucleic acid molecule and a first compound at the end thereof, and fixed on the substrate; an optical marker unit selectively combining to the first compound and retroreflecting emitted light; a light source unit emitting the light to the optical marker unit; and a light receiving unit receiving the light retroreflected from the optical marker unit. According to the optical gene biosensor of the present invention, accurate quantitative analysis of the target nucleic acid molecule can be performed for both a non-spectral light source and a spectral light source.
