The present invention relates to a novel nucleic acid probe for detecting 5S rDNA to perform FISH and an improved FISH method using the same and, specifically, to a nucleic acid probe for detecting 5S rDNA, providing efficiency of cost, time, and human resources and having high reproducibility and anti-resistance by having excellent hybridization efficiency, a component thereof, and a FISH method for detecting 5S rDNA using the same. According to the present invention, the nucleic acid probe is a universal probe capable of detecting various kinds of 5S rDNA loci from the plant to animal kingdom. Specifically, the nucleic acid probe is provided as probes for detecting 5S rDNA for an angiosperm, gymnosperm, and an animal, respectively. The nucleic acid probe provides a clear signal about minor loci even if performing FISH with other probes at the same time. The nucleic acid probe provides a background result without noise. According to the present invention, the FISH method reduces existing long FISH hybridization time up to 5 minutes at room temperature and does not require continuous input of skilled research human resources. The nucleic acid probe is useful for broad dielectric substance analysis.
