The present invention relates to a method for separating and manipulating micro particles via continuous phase modulation of a surface acoustic wave, the method comprising: a step (S100) of applying a standing wave to the fluid containing the particles to be separated, and finding Δf_optimal, a frequency difference between two waves at both sides, for moving the particles to be separated to one side while varying the frequency of the wave at one side among the waves generated at both sides; and a step (S200) of applying a normal surface acoustic wave to a channel by generating surface acoustic waves at both sides of the channel while the fluid, in which two or more kinds of particles including the particles to be separated are mixed therein, is contained in the channel so that the particles to be separated among the two kinds of particles mixed therein are transferred to one side of the channel wherein the normal surface acoustic wave is applied in such a manner that the frequency difference between the surface acoustic waves at both sides becomes Δf_optimal. Accordingly, it is possible to separate or manipulate particles on a large scale and to control the moving speed of the particles, and thus, the particles may be separated and manipulated in various forms.
