The present invention provides a method for generating spinal cord glutamatergic interneurons (V2a interneurons) from human pluripotent stem cells (hPSCs). In the method of the present disclosure, it is also possible to culture the first population of hPSCs in vitro in a neural induction medium comprising a retinoic acid signaling pathway activator, sonic hedgehog (Shh) signaling pathway activator and Notch signaling pathway inhibitor. Well, culture produces a second population of cultured cells that contain CHX10 + V2a interneurons.
