The present invention provides 3D cartilage organoid block prepared by differentiating mesenchymal stem cells into 3D spheroid cartilage tissue, basic unit for 3D cartilage spheroid block. The inventors have found that both the amount of GAG matrix and the expression of type II collagen have increased. Therefore, the method of the present invention provides a clinically applicable cartilage tissue by efficiently increasing the function of the cartilage differentiation construct by 2D culture. The 3D cartilage organoid block can be effectively applied to areas such as articular cartilage regeneration and plastic surgery where cartilage tissue recovery is required.
