In traditional approaches to plant breeding, chemical mutagenesis can be used to randomly introduce nucleotide substitutions into the plant genome, i.e. without the ability to control the sites of nucleotide changes. When it comes to finding a given nucleotide substitution, the statistical probability is extremely low due to the complexity of the genome. The present invention, however, has demonstrated how to develop a new alternative application of digital polymerase chain reaction (dPCR), preferably drip dPCR (ddPCR), to detect specific nucleotide substitutions in mutant genes. The entire platform includes a screening method with a library of mutagenized organisms, a system based on digital PCR, and an installation for reproducing and analyzing identified mutant organisms.
