摘要
The invention relates to a method for identification of bacterial genes involved in synthesis and metabolism of neuroactive compounds in microbiomes and individual bacterial genomes, in particular in the human intestinal microbiomes. It may be used in medicine, medical microbiology and neurology. The method uses a set of 45 oligonucleotide (primer) pairs for PCR-analysis of the total genomic DNA. The primers are selected for ten key genes coding the enzymes involved in synthesis and metabolism of neuroactive compounds: serotonin, dopamine, histamine, meatonin, gamma-aminobutyric acid, tyramine, spermidine, butyric acid, NO gas transmitter revealed in the genomes of 27 dominating bacterial species of the human intestinal microbiota. The developed primer pairs may be used for separate search for selected genes of neuroactive compounds in individual bacterial genomes and for monitoring a whole microbiome. The suggested sets of primer pairs may be used as diagnostic systems for investigating the microbiota under normal and pathological conditions. The primers are designed so that they may be used for determining a transcript level of single genes using a real-time PCR-analysis method.
