A method for inactivation and extraction of acid-resistant bacteria in a test sample, the method comprising the following sequential steps: (a) acquiring a test sample of a solid or semi-solid culture medium that is known to contain or that may contain acid-resistant bacteria and suspend the test sample in a container containing ethanol and beads; (b) treat the container with a beadbeater homogenizer to separate the small aggregates and / or break the cells of the acid-resistant bacteria in the container; (c) subsequently incubate the suspension for at least 10 minutes at room temperature to inactivate the acid-resistant bacteria contained in the test sample; (d) centrifuge the vessel to settle the sample of acid-resistant bacteria and remove the supernatant; (c) resuspend the sediment of acid-resistant bacteria in formic acid and (f) subsequently add acetonitrile to the container; wherein formic acid and acetonitrile extract and dissolve cellular proteins from inactivated cells.
