Protein enrichment method of a microalgae cultivated in heterotrophy, microalgae of the genus Chlorella, characterized in that the heterotrophic culture comprises a step that aims to limit the growth of said microalgae due to a lack of the glucose fermentation medium with a supply of glucose continuously in insufficient quantity in order to obtain a growth rate of the microalgae lower than that of said microalgae in the absence of nutritional limitation, and by an increase in the fermentation temperature with respect to the optimal fermentation temperature, allowing thereby achieving a protein content of the biomass greater than 50% by weight.
