The invention provides lipo-polymeric nanoparticles and their preparation. The process involves dissolving polydiacetylene in dimethyl sulfoxide and a phospholipid in chloroform followed by adding 1-Ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) and N-Hydroxysuccinimide (NHS) in 1: 1 ratio, at least one polyclonal or monoclonal antibody, 1-20 picomoles of a cationic molecule and keeping said sample overnight at 2-8°C, followed by keeping under UV light at intervals of 10 seconds for a total of 30-60 seconds to allow the polydiacetylene to polymerize and form lipo-polymeric nanoparticles. The invention also provides a method of detection of an analyte comprising contacting a sample with lipo-polymeric nanoparticles, wherein presence of an analyte such as Alpha-fetoprotein (AFP); and beta-human chorionic gonadotropin (HCG) in sample analysed by a shift in color from blue phase to red phase with an absorption maximum at about 540 nm.
