The present disclosure provides a technique which makes it possible to evaluate the state of a cell mass of one or more spheroids, etc. In this culture state determination device, a plurality of light sources sequentially illuminate a plurality of cell masses placed and held on an image sensor. The image sensor acquires a captured image of the plurality of cell masses every time the plurality of light sources illuminate the plurality of cell masses. At least one control circuit: extracts a region including an image of a cell mass in the captured image; generates three-dimensional image information of the region using a plurality of the captured images; extracts the external form of the cell mass and a cavity section inside the cell mass using the three-dimensional image information; defines a first volume, which is a volume based on the external form of the cell mass, and a second volume, which is a volume of the cavity section, on the basis of the external form and the cavity section of the cell mass in the three-dimensional image information; and determines a culture state of the cell mass using the first volume and the second volume.
