Provided is a detection method for tumour marker molecules, the method involving: first providing a multi-layered nano-pore array chip, which acts as a substrate; assembling the substrate and a liquid bath into a nano-pore sensor, wherein the nano-pore array chip is positioned at the centre of the liquid bath, and a solution at two sides of the liquid bath can only penetrate through the nano-pore array; anchoring antibody molecules on inner surfaces of silicon nano-pores on an intermediate layer of the substrate; adding tumour marker molecules on one side of the liquid bath; and finally, utilising electrophoresis to drive the tumour marker molecules to enter the nano-pore array. When the tumour marker molecules are captured by the antibody molecules in the multi-layered nano-pore array chip and a blocking current is generated, the concentration of the tumour marker molecules is determined according to a drop amplitude of the blocking current. The method is simple in operation, has excellent expansibility and is also widely applicable in the field of precise medical detection.
