Disclosed are a cultivation method for a supporting system of red plumage pink shell layer chickens, and a primer pair for identifying white leghorns red plumage mutagenic mutant genotype is provided. The primer pair consists of single-stranded DNA molecule shown by sequence 2 in the sequence table and single-stranded DNA molecule shown by sequence 3 in the sequence table; the primer pair designs a primer according to chicken reference genome Gallus_gallus-4.0 version sequence information No. 11 chromosome positive-sense strand 18,288,303rd deoxynucleotide upstream and downstream nucleotide sequence announced in NCBI. The disclosed method performs propagation on white leghorns of homozygous red plumage mutagenic mutant genotype by detection of the site genotype by means of restriction fragment length polymorphism, and breeds pure female parent; the hen filial generation of the female parent and Rhode Island Red male parent is 100% red plumage phenotype, which can meet market demands and has wide popularizing prospect.
