Disclosed in the present invention is a method for measuring an exonuclease activity in real time by an electrochemical measuring method based on a graphene electrode. The method for measuring an exonuclease comprises a step of making a sample which is expected to contain exonuclease react with double stranded DNA labeled with an electrochemical oxidation-reduction label; a step of making the resultant product of the sample and the double stranded DNA touch a graphene electrode; and a step of measuring a reductive peak current of the electrochemical oxidation-reduction label through the graphene electrode and detecting the exonuclease activity.
