Disclosed in the present invention are a PagSAP5 protein derived from Populus alba×Populus glandulosa for enhancing drought stress tolerance in a plant; a drought stress-tolerant gene encoding the protein; and a plant transformed by the gene and thus showing enhanced drought stress tolerance. The PagSAP5 gene/protein according to the present invention is involved in the tolerance to drought stress in a plant and thus may be beneficially used in the modification of fruit trees, crops, and the like by controlling the expression thereof. Furthermore, the plant transformed by the PagSAP5 gene shows excellent tolerance to drought stress, and thus may be beneficially used as a plant with increased biomass productivity in dry environments caused by climate changes caused by global warming and the like.
