The present invention relates to a composition for rapid and accurate detection of short nucleic acid molecules, and a detection method. More specifically, the present invention relates to a composition for miRNA detection, comprising: (a) a circular DNA template capable of being hybridized specifically to a target miRNA, and (b) a molecular probe including a nucleic acid material, a fluorescent material, and a fluorescence-reducing material, and a detection method of a target miRNA using the same. The present invention is a system capable of distinguishing the mutations in a single base sequence, such as single nucleotide polymorphisms (SNPs) and point mutations in a short miRNA, and also capable of detecting miRNAs existing in a trace amount in the body to attomole level. Furthermore, the detection method according to the present invention is simpler than a conventional method and can detect target miRNAs with higher sensitivity and accuracy within a short time.
