The present invention relates to a method of preparing the DNA library for a transposable element LINE-1(L1HS)-targeted probe enrichment using HiSeq sequencer, and more specifically, has developed an L1HS-targeted sequencing method on the basis of clinical and academic significance of transposable elements, the development of next-generation sequencing technology, cost reduction, and the development of analysis system. The present invention may serve as a method for use in studies of genetic variations due to L1HS in individuals and the genetic influences according to the same. Furthermore, based on the present invention, it is possible to further develop a technique capable of targeting L1HS-containing active transposable elements alone which have the ability to mobilize in the human genome and create genetic mutations. The transposable element-targeted sequencing as well as the targeted analysis sequencing method, as they are becoming simplified and commonplace, can be expected to be useful in the development of diagnostic markers in clinical medicine.
