The present invention relates to a method for separating and purifying high purity chlorophyll a from microalgae. More particularly, the present invention relates to a method capable of purifying chlorophyll a with a purity of 90% or more in a short time, wherein chlorophyll a is extracted from microalgae using ethanol and hexane, and then first purified using a column packed with Sephadex resin; and subjected to secondary purification using a column packed with C18 resin sequentially. Chlorophyll a purified by the separation and purification method of the present invention can be usefully used as a material for antioxidant cosmetics or functional health foods for antioxidants.
