The present invention relates to a novel real-time multiplexed multicolor BRET assay and device for studying protein-protein interactions and detecting diverse biological activities in living cells, wherein said assay comprises a single Samples and multiple signals from one or several wells at the same time. A real-time multiplexed multicolor BRET assay is used for drug screening, by altering the conformation of any channel or receptor subunit in living cells in response to protein interactions, potential drug candidates. It is particularly useful to indicate activation or inhibition of the receptor.
