PROBLEM TO BE SOLVED: To provide methods for easily determining the mechanism of action of an anti-inflammatory activity by an anti-inflammatory substance.SOLUTION: A method comprises: stimulating leukocytes or leukocyte-like culture cells responsible for innate immunity, with sarcoplasmic reticula/a sarcoplasmic reticulum CaATPase inhibitor, in a condition of Ca-free extracellular fluid; increasing the Caconcentration of the extracellular fluid in the presence or absence of an anti-inflammatory substance; measuring the intracytoplasmic Caconcentration and the superoxide production amount of the culture cells; and determining the Caentry pathway into the cytoplasm inhibited with the anti-inflammatory substance as a store-operated Ca entry pathway, when the intracytoplasmic Caconcentration and the superoxide production amount in the presence of the anti- inflammatory substance is reduced as compared with in the absence of the anti- inflammatory substance.SELECTED DRAWING: None
