The present invention provides an improved method for the production of CRM₁₉₇ with high yield using engineered Corynebacterium diphtheria strain having an increased copy number of the CRM₁₉₇ gene, wherein the method comprises growing the strain in media free of animal-derived components with one or more amino acids.
