The invention relates to a profile extraction method for 37 fatty acids derived from phospholipides, PLFA, used for the gas-chromatographic analysis of derivatized extracts. According to the invention, the method consists of 4 stages: saponification of soil sample with methanolic NaOH solution, at a temperature of 75...115°C, for 30 min, separation of phospholipid fraction from glycolipid and neutral lipid fractions, derivatization of the extract comprising the phospholipid fraction with a 1...6N solution of CHOH:HO with HCl, after which the extract is subjected to incubation at the temperature of 60...120°C, for 15 min, the derivatives of the fatty acids obtained thereby are further extracted with a mixture of organic solvents, by mechanical stirring, for 15 min, to result in an organic extract having a quality appropriate for gas-chromatographic analysis.
