OPLOPHORUS-DERIVED LUCIFERASES, NOVEL COELENTERAZINE SUBSTRATES, AND METHODS OF USE

An isolated polynucleotide encoding a modified luciferase polypeptide andsubstrate. The OgLucvariant polypeptide has at least 80% amino acid sequence identity to SEQ IDNO: 1 and comprising anaspartate, phenylalanine, glycine histidine, isoleucine, lysine, leucine,methionine, asparagine, proline,arginine, serine, valine, tryptophan or tyrosine at position 18 correspondingto SEQ ID NO: 1. TheOgLuc variant polypeptide has enhanced luminescence relative to thecorresponding polypeptide of thewild-type Oplophorus luciferase defined by SEQ ID NO: 3 in the presence of acoelenterazinesubstrate.
