Disclosed are a colorectal cancer solid tumour primary cell and colorectal cancer ascitic fluid primary tumour cell culturing method, and a matching reagent. The core of the present invention is: (1) using a mild cell dissociation reagent to treat colorectal cancer solid tumour tissue, and using a mild method to separate colorectal cancer cells in ascitic fluid, to ensure as far as possible the viability of the cancer cells; (2) preparing a special serum-free culture medium, and using a suspension culture system to perform in vitro culturing of tumour cells from a colorectal cancer solid tumour, to ensure cancer cell normal expansion while minimizing interference from normal cells. A colorectal cancer primary cell culture obtained by means of the present invention may be used for in vitro experiments at various cellular levels, secondary sequencing, animal model construction, cell line construction etc. It is expected that the culturing method has wide application prospects in the field of colorectal cancer research and clinical diagnosis and treatment.
