A composite microscope (10) employing two-photon excitation and stimulated emission depletion (STED) techniques, comprising a two-photon imaging unit (100) and an STED imaging unit (200). The two-photon imaging unit (100) can be used for a relatively thick sample, and the STED super-resolution imaging unit can be used for a region of interest in a surface layer of the sample, such that light spots generated after an excitation light and a depletion light are focused by an objective lens (OL) precisely overlap in a three-dimensional distribution.
