Disclosed in the present invention is a virtual PCR method for achieving sequence extension on the basis of NGS read searching. The method comprises: first performing one-time high-throughput higher-depth sequencing on an entire genome of a test sample on the basis of an NGS sequencing technique to obtain massive sequencing reads that cover the entire genome; then in combination with a bioinformatics method, searching the sequencing reads. Sequence extension is achieved in a program according to the search result, thereby establishing a contig of sequence fragments, and further achieving the amplification of a target gene in a virtual program.
