The preparing method comprises conducting an enzymatic reaction system containing uridine or cytidine solution, E. coli cell crushing solution with the activity of uridine/cytidine kinase and polyphosphate kinase, sodium hexametaphosphate, magnesium sulfate and a small amount of ATP. The reaction is performed under the conditions of pH of 8.5 and 32°C to synthetize UMP or CMP. In the above coupled catalytic reaction system, uridine/cytidine kinase is responsible for catalyzing uridine or cytidine and ATP to produce UMP or CMP, accompanied by dephosphorylation of the ATP to form ADP. The polyphosphate kinase is responsible for catalyzing sodium hexametaphosphate and ADP to form ATP, thereby realizing the regeneration of the ATP in the reaction. The method for preparing UMP and CMP provided by the present invention has the advantages of low raw material price, short cycle, simple operation, green environmental protection and high yield, and has good industrial application value.
