The present invention provides a method for rapidly and accurately analyzing polyoxin D remaining in fresh agricultural products with simple preprocessing by using a high performance liquid chromatograph-mass spectrometer (HPLC-MS/MS). An analysis method known so far requires an analysis time of 1 to 2 weeks, such as culturing microorganisms, by a microorganism assay (reverse valence assay) method of examining the degree of growth inhibition of bacteria by adding agricultural chemical components to a medium in which test bacteria are cultured, and the accuracy of quantitative analysis of a test results is low. According to the present invention, 10 g of an analysis sample consisting of homogenized fruits or vegetables is weighed in 50 mL of a centrifuge tube during preprocessing, 10 mL of 1% formic acid-containing methanol (MeOH) is added thereto, and a mixture is agitated for 15 minutes for extraction. When the analysis sample is a dry sample, 10 mL of water is mixed with 5 g of the dry sample and left for 1-4 hours, 10 mL of 1% methanol (MeOH) is added thereto, and a mixture is agitated for 15 minutes for extraction. After the extraction, 1 mL of extract is used for a solution for analysis after contrifuging the extract for 5 minutes at 4,000 rpm/min, and analysis can be performed in a short period of time within 10 minutes with the HPLC-MS/MS.
