Gene editing using modified closed terminal DNA (CEDNA)

This application describes ceDNA vectors with linear and continuous structures for gene editing. The ceDNA vector includes an expression cassette flanked by two ITR sequences, where the expression cassette encodes a gene editing molecule. Some ceDNA vectors further include cis-regulatory elements, including regulatory switches. Methods and cell lines for reliable gene editing using ceDNA vectors are further provided herein.
