A bilayer membrane containing sodium alginate/gelatin coated with electrospun polycaprolactone/gelatin/β-ginseng tricalcium phosphate nanopowder on one surface was synthesized and evaluated for bone regeneration application. The main object of the present invention is to produce a useful composite membrane that not only induces bone regeneration, but also has sufficiently strong tensile strength and prevents soft tissue migration from bone defects. The role of the SA/gel-coated surface in the membrane is that the porous electrospinning layer supports bone cell growth and promotes bone regeneration activity, while improving wound covering and preventing soft tissue migration. Cell viability and cell proliferation were obtained by MTT analysis and SEM images of MCT3T3 and L929 cells. The effects were compared at 2 weeks and 4 weeks with or without bilayer membrane treatment on rabbit radius bones. As a result of observation by micro-CT reconstructed images by N-recon software, the bilayer structured film showed good mechanical strength and bone induction after 4 weeks.
