The present invention relates to a method for expressing daidzein reductase, dihydrodaidzein racemase, dihydrodaidzein reductase, and tetrahydrodaidzein reductase, which are enzymes derived from Slackia isoflavoniconvertens, and for synthesizing equol or an equol derivative in a position selective and efficient way by using recombinant Escherichia coli in which the tetrahydrodaidzein reductase includes a mutant. In addition, the equol derivatives such as synthesized equol, 5-hydroxy-equol, 6-methoxy-equol, and ortho-hydroxide equol (3-hydroxy-equol, 6-hydroxy-equol,-8 hydroxy-equol) are female hormone analogues, and can be used as health function food for relieving menopausal symptoms. Moreover, the equol derivatives can be applied as functional ingredients of a cosmetic product having anti-oxidant and anti-aging effects. In particular, equol can be used as a relieving and therapeutic agent for ovarian cancer and prostate cancer while 5-hydroxy-equol can be used as a therapeutic agent for liver cancer.;COPYRIGHT KIPO 2020
