The present invention relates to an in vitro cell-free expression system comprising a novel inorganic polyphosphate-based energy regeneration system. In a specific embodiment, the present invention includes a cell-free expression system in which ATP, a cellular energy source, is generated from an inorganic polyphosphate using a dual enzyme system. In this embodiment, the dual enzyme system may include a thermostable adenosyl kinase, and/or a polyphosphate kinase enzyme.
