FIELD: biotechnology.;SUBSTANCE: invention refers to biotechnology and molecular biology. Disclosed is a method of determining a change in a subject's immune repertoire, comprising steps of quantitative measurement of immune cell clenotypes in a population or subpopulation of immune cells in a first sample and a second sample taken from one patient, wherein clones of immune cells are quantitatively measured using polynucleotide extraction, arm-PCR and subsequent generation sequencing, calculating frequency data for the first sample and the second sample by identifying the frequency of each clonotype, normalizing the frequency data for the first sample and the second sample, determining the absolute difference in frequency of each clonotype present in the first sample and the second sample, and determining common clonotypes for the first sample and the second sample with the highest frequency variation.;EFFECT: method can be used in medicine due to accuracy of measurement of metabolic rate of the immune repertoire and possibility to classify T- and B-lymphocytes at the level of one cell.;16 cl, 5 dwg, 2 tbl, 3 ex
