A manifold 111 for a medicament delivery device 100 to deliver discrete medicament doses contained in a plurality of compartments 118 spaced along a carrier strip 120 and sealed by a sealing layer 122 to form a medicament carrier 116. Manifold 111 comprising: an opening mechanism comprising a release member 134 to engage between carrier-strip 120 and sealing-layer 122 to separate them from each other, thereby opening a compartment 118 as medicament-carrier 116 advances to an entrainment position. Release-member 134 includes a surface that covers the opened compartment to substantially contain the medicament therein when the compartment’s in the entrainment position. Preferably release member 134 forms part of an entrainment airpath by providing an inlet and an outlet for air to pass through the opened compartment via the release-member’s compartment covering surface to entrain the medicament. The opening mechanism may comprise an indexing wheel 126 with compartment receiving indentations, where rotating indexing-wheel 126 moves compartments 118 sequentially into the entrainment position. Also a medicament delivery device 100 comprising a manifold 111, device 100 preferably comprising a tensioning mechanism to alter a path length that the separated sealing-layer 122 travels through device 100 to regulate tension in the separated sealing-layer 122.
