A composition of plasmids comprising a first plasmid coding for the genome of measles virus comprising a single additional transcriptional unit (ATU) and a helper plasmid coding for and expressing the N, P and L proteins of measles virus, and pharmaceutically acceptable excipients; wherein the ATU comprises two or more non-viral genes, such that the ATU is bicistronic, tricistronic or tetracistronic. The ATU may code for combinations of GMCSF, cytosine deaminase, soluble PD-1 and prostate specific acid phosphatase. The plamids may be used to produce a recombinant measles virus for treating cancer.
