Toxicity Studies on Nsc 109229, L-Asparaginase, Merck, Sharp and Dohme, C-7956, Lot No. 0965-Hls-2-028, Following Daily I.V. Administration to a Rhesus Monkey

A single Rhesus monkey was treated with a dosage schedule of 1,000 I.U./kg/ day x 5, I.V. and sacrificed seven days post Rx. During the treatment period the animal showed a weight loss which was associated with a decrease in food consumption. Drug treatment resulted in a fatty liver which was seen both on gross and histological examination; BSP retention which was first noted one hour after 3,000 I.U./kg and which continued through the treatment and observation periods but was markedly improved on the day of sacrifice; a slight anemia which was noted on the day of sacrifice which we attribute to the multiple bleedings; no marked change in the leucocyte count during the course of the study nor any significant alterations in the serum proteins or cholesterols; a suggestion of a delay in prothrombin time on the fifth and eighth days of the study but not on the twelfth; serum triglycerides were not elevated on the fifth day of the study but there was a slight increase on the twelfth day. Urinalyses done during the study were not significantly different than those obtained during the pretreatment period. (Author)
