Emulphor (EL-620) Vehicle: Blood Pressure Effects of I.V. Infusion in Dogs and Rhesus Monkeys.

Emulphor (EL-620), a possible vehicle for I.V. infusion of CCNU (NSC-79 037) and Methyl CCNU (NSC-95 441),was tested for blood pressure changes in 3 unanesthetized dogs and 2 unanesthetized monkeys. Treatments were performed with saline containing 5% Emulphor and 5% Ethanol. The rapid I.V. injection of 10mg/kg Emulphor (0.2ml/kg) produced a severe decrease of blood pressure,with bradycardia and severe sinus arrhythmia. The infusion of 187.5mg/kg Emulphor (3.75ml/kg,within approximately 60minutes) produced a similar drop in blood pressure. Fifteen to 30minutes after onset of treatments there also occurred erythema and edema of the ears,injection of the conjunctivae and hyperemia of the eyelids,which lasted approximately 60minutes. Pretreatment with the above rapid I.V. injection as a priming dose,reduced the blood pressure effects of the subsequent Emulphor infusion. The 2monkeys that were infused I.V. for 60minutes with Emulphor 666.0mg/kg (13.3ml/kg) showed a slight decrease of blood pressure,which was less than 20mm Hg,without any other toxic manifestations.
