An Abbreviated Toxicologic Evaluation of Three Platinum Containing Compounds -NSC-119,875,NSC-241,240,NSC-271,674in Dogs.

Three platinum containing compounds (NSC-119,875,NSC-241,240,NSC-271,674) were given as a single intravenous dose to beagle dogs. The lowest emetic doses were 9mg NSC-119,875/sq. m. (0.45mg/kg), 624mg NSC-241,240/sq. m. (31.2mg/kg) and 46mg NSC-271,674/sq. m. (2.3mg/kg). All three compounds caused emesis,anorexia,decreased food intake and weight loss. Leukopenia occurred following treatment with each of the compounds. Thrombocytopenia was noted in dogs given either NSC-119,875or NSC-241,240. A mild decrease in the hematocrit,hemoglobin and erythrocytes occurred in female dogs only. Although all three compounds caused this change,the incidence was greatest with NSC-119,875. Histopathologic lesions included renal tubular degeneration and hypocellularity of the bone marrow for all three compounds. NSC-241,240also caused an elevated M/E ratio and NSC-271,674caused a mild lymphoid depletion of the Peyer's patches. Although NSC-241,240had the lowest emetic potential,it induced several of the lesions described above at doses less than its lowest emetic dose,whereas neither NSC-119,875nor NSC-271,674caused any significant changes at a dose less than that required to cause emesis.
