Techniques to effectively labeling blood cell specific monoclonal antibodies (MAbs), with In-111 using bifunctional chelating agents such as the cyclic or mixed anhydride of diethylenetriaminepentaacetic acid have been developed. Conditions for the optimal labeling of blood cells are reported. The specificity (Kd values) of In-111 labeled MAbs for the cell specific antigens are reported. In-111-MAb labeled canine platelets were used to image vascular thrombi. Under the best experimental conditions approximately 70% of the radiolabeled MAbs get incorporated into isolated blood cells. However, only about 35% of the added MAb-associated radioactivity would incorporate into the desired type of blood cells in whole blood. The chemical nature of the radioactivity that does not bind to the cells was investigated. 9 figs., 6 tabs. (ERA citation 12:019165)
