Influenza viral genetics – Continued studies of temperature sensitive (ts) mutants have been productive in more precise definition of the genetic lesion with respect to viral function.nB.tApplication of viral genetics to improvement of influenza vaccines – We have continued to update both high yield conventional bi-specific vaccine candidate strains and antigenically hybrid neuraminidase specific vaccine strains by recombination as new wild type variants have appeared.nC.tIsolation and characterization of influenza viral polypeptides – We can now resolve and isolate each of the 5 major influenza viral polypeptides through combination of detergent disruption of the particle, affinity chromatography and gel electrophoresis.nD.tStudies of influenza viral replication and its inhibition - We have continued to make use of the synthetic neuraminidase substrate 2-3 methoxyphenol-N- acetylneuraminic acid (MPN) in studies of influenza virus replication and function.
